Talk title: Cracking the Code: Molecular basis of Species-Specific Implantation Revealed Through Embryo Models.

Abstract: The implantation of mammalian embryos into the uterus is a critical process establishing the first connection between competent blastocysts and maternal tissues necessary for nutrient exchange and waste removal during gestation. During implantation, the blastocyst first interacts with the uterine tissue and then later invades it. However, the molecular mechanisms regulating these physiological adaptations remain largely elusive.

In this study, we constructed trophectoderm-specific gene regulatory networks using stem cell based embryo models, blastoids. We integrated machine learning algorithms to predict the effects of gene perturbations, and combined these predictions with functional analysis of CRISPR-based gene knockouts. By establishing a molecular hierarchy among these transcription factors, we elucidate their interplay during early implantation. Additionally, deep learning models of chromatin accessibility offered insights into the evolutionary adaptations of genomic elements that regulate crucial gene modules in a spatiotemporal manner. These findings suggest potential strategies employed by different mammalian species to ensure reproductive success and survival in diverse environments.

This talk will also highlight the immense potential of human blastoid and endometrial organoid systems. These models provide powerful platforms to investigate maternal-foetal interactions, offering novel insights into the complex dynamics of embryonic development and high rate of pregnancy failures in humans compared to other species.
